Case Studies

The following applications have been successfully tested with syntrix or noviogel:

Case 1: syntrix-BM: tumor co-culture

Syntrix-BM: 20k PC346c +10k CAFs
BME: 20k PC346c +10k CAFs

Syntrix-BM is a fully synthetic, biomimetic matrix displaying cellular binding sites mimicking important ECM components present in laminin rich ECM gels, thereby offering a more defined alternative. Here, we present a coculture model of the PC346c prostate cancer cell line labeled with RFP and Cancer Associated Fibroblasts (CAFs) labeled with GFP. Varying concentrations of CAFs were used to investigate the crosstalk between cancer and stromal cells. Both fluorescent labels are stably expressed. Confocal images taken after 21 days in culture show that PC346c cells and CAFs are both still present. The PC346c cells form tumor spheroids, while the CAFs are interspersed with the PC346c clusters. At the higher concentrations, some clustering of CAFs with PC346c cells occurs in BME. (for more information see application note below)

Case 2: syntrix-BM: cellular imaging and analysis


Here we show that fully synthetic, biomimetic Syntrix matrices displaying five peptides derived from laminin, fibronectin, and collagen IV. Syntrix-BM provides an excellent cell-instructive 3D cell culture environment for encapsulated cells.The liver, as a primary organ for drug metabolism, is the main target of drug toxicity. For this reason, many in vitro cellular cytotoxicity studies focus on liver toxicity. The HepG2 human hepatoma cell line is a common cell line used in hepatotoxicity models. We demonstrated the feasibility of live-cell imaging by monitoring cellular response after a toxic insult with acetaminophen (AAP) using different fluorophore dye mixtures.cells incubated with 0, 1, 10, 20, 100 and 200 mM AAP and stained with MitoTracker Green and HOECHST. Specific mitochondrial staining of these powerful dyes is clearly visible in the control and concentrations up to 20 mM. The mitochondria can be distinguished as bright green dots in the cytoplasm of the cells. The staining indicates that 100 and 200 mM AAP have profound effects on cell viability as with 100 mM AAP the cytoplasm displays a faint green stain losing most of the granular structure and with 200 mM no MitoTracker dye has been taken up by the mitochondria anymore, clearly demonstrating the usefulness of the Syntrix-BM Kit for cytotoxicity measurements. Case 3: noviogel-RGD: Maturation of follicles into oocytes


Successful reproduction in males and females is vital to survival and is dependent on several factors including development of reproductive organs.Ovarian development in mammals can be split into two phases, 1) growth of the ovary and establishment of the founding primordial follicle pool and 2) maturation of follicles and release of a fertilizable egg starting at puberty. The figure shows the maturation of follicles into oocytes in noviogel-RGD and Matrigel.

Case 4: noviogel: HUVECs sprout with RGD linked to noviogel


HUVECs were grown in fibrin gel, noviogel-P and noviogel-RGD. The fibrin served as a control and showed as expected sprouting from day 8 till day 21, were it shows a more pronounced vascular network. Noviogel-P showed no sprouting because of the lack of attachment sites. By coupling a RGD peptide to noviogel resulted in sprouting and a vascular network from day 8 till more pronounced at day 21 (figure 1). In the second figure, the proliferation of the HUVECs is shown with the effect of growth factors on the prolferation of HUVECs. Adding growth factors increased the growth of HUVECs inside the hydrogel. The last four bars show the difference between growth factors coupled to the hydrogel or added to the medium as free molecules. No significant difference was observed between the two groups. Case 5: noviogel-RGD: Dog primary liver organoids


Dog primary liver cells were cultured in Matrigel and noviogel-RGD for 7 days. Liver organoids cultured in noviogel-RGD showed excellent growth characteristics, more organoids and bigger in size.
